In recent years, various optical clearing techniques for fixed tissues of mammalian brain have been actively adopted to know details of brain structure including neuronal circuits. Here we describe the outline of ScaleS method, a tissue-clearing technology we developed, by exhibiting of the application. Our three-dimensional tissue-staining techniques compatible to ScaleS-conducted clarification of brain tissues, namely AbScale method (immunostaining) and ChemScale method (chemical dye-staining), are also introduced with their examples of Alzheimer disease pathology and neural stem cell biology in this article.
