Amyloid hypothesis and tau hypothesis

The long-standing “amyloid hypothesis” that Alzheimer's disease is cause by the production and aggregation of amyloid-beta faces serious challenges by data recently obtained from neuroimaging studies and amyloid-beta-focused clinical trials.  In this review, we inspect a problem of amyloid hypothesis and propose a new hypothesis based on accumulating evidences and new findings.
