NSAIDs and Alzheimer's disease

Alzheimer's disease (AD) is the most common neurodegenerative disorder of the central nervous system and the leading cause of adult onset dementia. Some epidemiological studies suggested that use of non-steroidal anti-inflammatory drugs (NSAIDs) suppresses the onset of AD. Various mechanisms, such as anti-inflammation and γ-secretase modulation were proposed to explain this effect of NSAIDs. Unfortunately, several clinical trials that tested the efficacy of NSAIDs for AD produced negative results. In this article, we review the basic studies, epidemiological studies and the clinical studies for the effect of NSAIDs on AD.
